MT-1 (Melanotan-1 / Afamelanotide) is a synthetic linear analogue of alpha-melanocyte stimulating hormone (α-MSH), differing from the native peptide by a single amino acid substitution (norleucine for methionine at position 4) which confers enhanced receptor binding affinity and metabolic stability. It acts as a potent MC1R agonist and is studied for its roles in melanogenesis, UV-response pathways, and photoprotection biology.

MT-1 has been the subject of both preclinical and clinical research. The EMA-approved form (afamelanotide) is used clinically for erythropoietic protoporphyria — making this one of the few melanocortin research peptides with a regulatory approval precedent. Preclinical research explores MC1R-mediated signalling, pigmentation pathways, immune modulation, and inflammatory response.
