How to Reconstitute Peptides: Complete Step-by-Step Guide
This guide covers everything you need to know about reconstituting lyophilised (freeze-dried) research peptides safely and accurately. Whether you are working with BPC-157, TB-500, GHK-Cu, or any other research peptide, the principles remain the same.

Why Bacteriostatic Water?
Bacteriostatic water (BW) is the standard reconstitution solvent for research peptides because:
- The 0.9% benzyl alcohol acts as a preservative, inhibiting microbial growth in the reconstituted solution
- Reconstituted peptides stored in bacteriostatic water typically remain stable for 4-6 weeks when refrigerated at 2-8°C
- Sterile water without preservative should only be used for single-use preparations, as it provides no microbial protection
Alternative solvents: Some peptides (e.g. those with low water solubility) may require 0.1-1% acetic acid or DMSO as an initial solvent before dilution with bacteriostatic water. Check the individual product data sheet or CoA for guidance.
Step-by-Step Reconstitution Process
Step 1: Prepare Your Workspace
Work in a clean area. Wipe your work surface with an alcohol swab and allow it to dry. Wash your hands thoroughly or use nitrile gloves throughout the process.
Step 2: Calculate Your Volume
Decide how much bacteriostatic water to add to your peptide vial. The volume you choose determines the concentration of your solution.
Example: If you have a 5mg peptide vial and add 1mL of bacteriostatic water, your concentration is 5mg/mL (5,000mcg/mL). If you add 2mL, your concentration is 2.5mg/mL (2,500mcg/mL).

Step 3: Sterilise the Vial Stoppers
Using a fresh alcohol swab, wipe the rubber stopper of both the bacteriostatic water vial and the peptide vial. Allow them to air dry for 30 seconds before proceeding.
Step 4: Draw Bacteriostatic Water
Using your insulin syringe, draw the calculated volume of bacteriostatic water from its vial. For example, to add 1mL to your peptide vial, draw 1mL (100 units on an insulin syringe).
Step 5: Inject Slowly - Aim for the Vial Wall
This is the most critical step. Insert the needle into the peptide vial at an angle and slowly inject the bacteriostatic water down the side of the vial, not directly onto the peptide cake. Direct injection can cause foaming and degrade the peptide.
Inject slowly over 10-20 seconds.
Step 6: Allow to Dissolve
Most peptides will dissolve within 1-5 minutes. Do not shake the vial - shaking can cause foaming and peptide degradation. Instead, gently swirl or roll the vial between your palms. If the peptide has not dissolved after 10 minutes, allow it to sit at room temperature for up to 30 minutes.
Step 7: Inspect the Solution
Once dissolved, the solution should be clear and colourless (some peptides produce a very slight yellow tint). If the solution is cloudy, contains visible particles, or has not dissolved after 30 minutes, contact us before proceeding.
Storage of Reconstituted Peptides
- Refrigerate at 2-8°C immediately after reconstitution. Never freeze a reconstituted peptide solution.
- Protect from light. Store vials in a dark location or wrapped in foil.
- Use within 4-6 weeks. Reconstituted peptides in bacteriostatic water are typically stable for 4-6 weeks when properly refrigerated.
- Lyophilised (unreconstituted) peptides should be stored at -20°C for long-term storage and at 2-8°C for short-term storage (up to 3 months).
Dosage Calculation
Use this formula to calculate how many units to draw from your insulin syringe:
Volume to draw (units) = (Desired amount in mcg ÷ Total peptide in vial in mcg) x Total volume added (mL) x 100
Example:
- Peptide vial: 5mg (5,000mcg)
- Bacteriostatic water added: 1mL
- Desired amount: 250mcg
- Calculation: (250 ÷ 5,000) x 1 x 100 = 5 units on the insulin syringe

Common Questions
Can I use regular sterile water instead of bacteriostatic water?
Regular sterile water (without benzyl alcohol) can be used for single-use preparations, but it provides no microbial protection. Reconstituted solutions in plain sterile water should be used immediately and not stored. Bacteriostatic water is strongly preferred for research requiring multiple draws from the same vial.
My peptide is not dissolving - what should I do?
Some peptides have low water solubility. Try adding a small amount of 0.1% acetic acid (10-20% of your final volume) to the vial first, allow the peptide to dissolve in this, then dilute to your desired concentration with bacteriostatic water. If problems persist, contact our team.
How do I know if my peptide has degraded?
Signs of peptide degradation include: cloudiness in a previously clear solution, visible particles, unusual colour changes, or a strong or unusual odour. When in doubt, discard and reconstitute a fresh vial.
What syringes should I use?
Insulin syringes (U-100, 1mL) are the standard choice for research peptide work. They are calibrated in units (100 units = 1mL), making dosage calculation straightforward. We stock BDu100 31g syringes suitable for research use.
Does bacteriostatic water expire?
Yes. Check the expiry date on your bacteriostatic water before use. Using expired bacteriostatic water may compromise peptide stability and research integrity.
