ARA-290 (Cibinetide) is an 11-amino-acid linear peptide (Glp-Glu-Gln-Leu-Glu-Arg-Ala-Leu-Asn-Ser-Ser; CAS 1208243-50-8) engineered from the aqueous face of helix B of erythropoietin (EPO). The sequence corresponds to a region of the EPO tertiary structure that is not involved in binding the classical homodimeric EPO receptor, allowing ARA-290 to engage the innate repair receptor (IRR) — a heteromeric complex of the EPO receptor and the β-common receptor (CD131) — without the erythropoietic activity of the parent hormone.
Because it dissociates tissue-protective signalling from haematopoietic signalling, ARA-290 is a widely used research tool for isolating IRR-mediated pathways. The research literature examines its role in models of neuropathic pain, small nerve fibre density, sarcoidosis-associated neuropathy, ischaemia-reperfusion injury, and inflammatory resolution, as well as its pharmacokinetic profile relative to full-length EPO.
