Glutathione (GSH) (γ-Glu-Cys-Gly; CAS 70-18-8) is a tripeptide present at millimolar concentrations in most mammalian cells. Its defining structural feature is the gamma peptide bond linking the glutamate side-chain carboxyl to cysteine, which renders it resistant to cleavage by standard intracellular peptidases. The free cysteine thiol is the reactive centre responsible for its redox chemistry.
Glutathione is the principal intracellular redox buffer, cycling between its reduced (GSH) and oxidised disulfide (GSSG) forms. It is a standard reagent and reference compound in research on reactive oxygen species scavenging, glutathione peroxidase and glutathione S-transferase enzymology, xenobiotic conjugation and phase II detoxification, and the GSH/GSSG ratio as a marker of cellular redox state. Biosynthesis proceeds via glutamate-cysteine ligase (the rate-limiting step) and glutathione synthetase, with regeneration from GSSG catalysed by NADPH-dependent glutathione reductase.
