Ipamorelin (Aib-His-D-2-Nal-D-Phe-Lys-NH₂) is a synthetic pentapeptide and the first truly selective growth hormone secretagogue (GHS), developed as a tool to distinguish GHRP-mediated GH release from off-target ACTH, cortisol, and prolactin stimulation seen with earlier GHRPs such as GHRP-2 and GHRP-6. It acts as a potent, selective agonist at the ghrelin receptor (GHS-R1a).
Ipamorelin's exceptional GH-specificity makes it particularly valuable in research on the ghrelin/GHS-R1a axis, pituitary somatotroph function, and GH/IGF-1 pathway modulation without the confounding endocrine effects of less selective GHRPs. Pharmacokinetic studies in humans have characterised its dose-proportional GH stimulation profile, short half-life (~2 h), and clean endocrine selectivity profile.
