KPV (Lys-Pro-Val; α-MSH residues 11–13) is the C-terminal tripeptide of alpha-melanocyte stimulating hormone (α-MSH). While short, KPV retains the principal anti-inflammatory bioactivity of the parent 13-residue peptide, acting primarily via MC1R-mediated pathways to suppress NF-κB activation and pro-inflammatory cytokine release (TNF-α, IL-1β, IL-6) in immune and epithelial cell models.
Research has demonstrated KPV's anti-inflammatory efficacy in models of colitis, skin inflammation, and wound healing. Its small molecular size, resistance to proteolysis (conferred by the C-terminal valine), and ability to modulate both innate immune and adaptive inflammatory responses make it a highly versatile research peptide. It has been explored for delivery via oral and topical routes in preclinical models.
