Semax (Met-Glu-His-Phe-Pro-Gly-Pro) is a synthetic heptapeptide analogue of the ACTH(4-7) fragment, with a C-terminal Pro-Gly-Pro extension that confers improved metabolic stability over the parent peptide. Developed originally in Russia, it is widely studied for its role in BDNF (brain-derived neurotrophic factor) upregulation, neuroprotective signalling, and modulation of the hypothalamic-pituitary-adrenal (HPA) axis.

Preclinical research has investigated Semax in stroke models, ischaemia-reperfusion studies, cognitive function, and memory consolidation pathways. Its ACTH-derived structure enables interaction with melanocortin receptors (MC2R/MC4R), while the Pro-Gly-Pro extension increases peptidase resistance versus the parent ACTH(4-7) fragment.
