SLU-PP-332 is a potent pan-agonist of the estrogen-related receptor (ERR) family — ERRα, ERRβ, and ERRγ — developed at St Louis University as a selective chemical probe for ERR-mediated transcriptional programmes. It functions as an exercise mimetic by activating the same mitochondrial biogenesis and oxidative metabolism pathways induced by physical exercise, without requiring physical activity.

Mechanistically, SLU-PP-332 drives ERR-dependent transcription of genes involved in fatty acid oxidation, OXPHOS complex assembly, and mitochondrial fusion dynamics. In rodent studies, it has been shown to increase aerobic exercise capacity and activate skeletal muscle fibre type switching. Its selectivity for ERRα/β/γ over classical oestrogen receptors (ERα/β) makes it a valuable tool for dissecting ERR-specific biology.
