TB-500 (889) is a synthetic version of the actin-sequestering active region of Thymosin β4, corresponding to the LKKTETQ heptapeptide motif (amino acids 17–23 of full-length Thymosin β4; parent protein CAS 107761-42-2). The 889 designation refers to the specific fragment studied extensively for its role in cellular migration, angiogenesis, and tissue-response pathways.
Research has established that the LKKTETQ actin-binding domain is primarily responsible for the wound-healing and angiogenic activity observed with full-length Thymosin β4. TB-500 sequesters G-actin, reducing the intracellular G/F-actin ratio and thereby promoting cell motility, blood vessel formation, and matrix remodelling.
